Jujube seed extract powder is obtained from the dried core and inner kernel of the mature seeds of Ziziphus jujuba var. spinosa, a species within the Ziziphus genus of the Rhamnaceae family. This extract is recognized as a traditional Chinese medicine in the Chinese Pharmacopoeia. The saponins derived from Ziziphus seeds deliver various pharmacological benefits, including calming and sleep-promoting effects, blood pressure reduction, protection against myocardial ischemia, cholesterol lowering, atherosclerosis prevention, brain safeguarding, and resistance to lipid peroxidation. These saponins fall under the category of triterpenoid tetracyclic compounds, primarily featuring components such as jujubogenin A, jujubogenin B, jujubogenin B1, jujubogenin E, jujubogenin D, and jujubogenin H.

Anti-anxiety and anticonvulsant: It can counteract seizures triggered by substances such as strychnine and pentetrazol, while also lowering mortality rates.

Analgesic and fever-reducing: The extract provides significant pain relief and can lower body temperature in normal mice, with effects lasting extended periods.

Antiarrhythmic properties: Particularly effective in preventing and treating arrhythmias caused by aconitine.

In rats on a high-fat diet, it significantly reduces serum triglycerides (TG) and boosts HDL, helping to inhibit atherosclerosis development through lipid and lipoprotein regulation.

Anti-platelet aggregation: The extract's oil component effectively blocks ADP-induced platelet clumping in mice.

Alcoholic extracts of the seeds improve the phagocytic activity of mouse mononuclear macrophages, stimulate lymphocyte transformation, and boost antibody production.

The primary approach for extracting saponins from jujube seeds involves solvent-based methods. To enhance extraction efficiency, supplementary techniques such as semi-bionic extraction, enzyme-assisted extraction, microwave extraction, ultrasonic extraction, or high-pressure extraction are often employed for processing raw saponins. However, the purity of saponins from these methods typically ranges from 5.3% to 18.3%, which can limit their applications. Therefore, advanced extraction and purification processes are essential for ensuring safe and effective use.

The following outlines the procedure for preparing jujube seed extract:

(1) Place Ziziphus spinosa seeds in a low-vacuum dryer with cool air and maintain at 50°C for 90 minutes to dry the seed exterior and facilitate easier grinding.

(2) Grind the dried seeds into a fine powder, then mix the powder with water to form a 12% (w/w) suspension.

(3) Heat the suspension to 56°C at a natural pH for 2.2 hours in a sealed device, introducing high-pressure steam from the bottom while stirring for 42 minutes to create a prepared liquid.

Evacuate the device using a vacuum system during stirring to eliminate any bubbles.

(4) Transfer the liquid into a ball mill with agate balls, grind for 1.5 hours to avoid degrading active ingredients, then heat to 70°C and hold for 2 hours to ensure stability and quality. Follow with ultrafiltration, homogenization at 20 MPa pressure, sieving through a 200-mesh filter, and collect the resulting liquid.

(5) Add the filtrate to a reactor, stir continuously for 18 minutes at 250 rpm, divided into the following stages:

Stage 1: Maintain reactor pressure at 130 mbar and temperature at 55°C for 3 minutes.

Stage 2: Set pressure to 110 mbar and temperature to 52°C for 3 minutes.

Stage 3: Adjust pressure to 95 mbar and temperature to 49°C for 3 minutes.

Stage 4: Fix pressure at 90 mbar and temperature at 48°C for 3 minutes.

Stage 5: Set pressure to 85 mbar and temperature to 47°C for 3 minutes.

Stage 6: Adjust pressure to 75 mbar and temperature to 46°C for 3 minutes.

This process ensures even distribution of the powdered material, maximizing therapeutic benefits, improving uniformity, and enhancing digestibility and absorption.

(6) Dry the mixture and grind the dried material into a fine powder to yield the final jujuboside sample.

For dispersible tablets of Ziziphus saponins, the preparation involves:

1) Measure 5.0 g of Ziziphus saponin extract, 3.0 g croscarmellose sodium, 10 g microcrystalline cellulose, 1.0 g talc, and 0.5 g stevioside, then pass through an 80-mesh sieve.

2) Use Ziziphus seed saponin extract as the base, microcrystalline cellulose as the filler, croscarmellose sodium as the disintegrant, stevioside as the sweetener, and 75% ethanol as the binder. Granulate through an 18-mesh stainless steel sieve, dry the wet granules in an oven at 50-60°C for 1-2 hours, and then sieve the dry granules through an 18-mesh sieve.

3) Mix in the talc, blend uniformly, and compress into tablets to produce 50 dispersible tablets of total Ziziphus saponins.
